Wedo5%silaneineitherabsoluteethanolorin95%ethanol(5%H²0). Watercatalyzesthesilanizationandsoyouneedtoreactabitlongerwiththeanhydrousethanoltogetgoodcoverage.Typicallyforanhydrousconditions,wereactfor~30minutesandforthe5%aqueous,wereactonly10minutes.Wedidnotfullycharacterizethesurfacecoverageofthefunctionalgroupsonthesurface. YoucouldtakethedeviceapartanduseXPStolookattheatomiccompositionofthesurface. Wedeterminedtheoptimalconditionsmoreempiricallythanthat-wemodifiedconditionsforthehighestdensitysurfacecoveragebythecellsthatwewantedtocapture.Ken

ItwasneutralpH,butcanandperhapsshouldbeslightlyacidic. Youcanmaketheethanolsolutionslightlyacidic~pH5 withaceticacid. Thiswillspeedupthereaction,andyouwillneedtoshortenthetimeinsilane/ethanolsolutiontoabout5minutes.Alsobesuretowashthroughlyifyougothisroute.

Ihavefoundyourworkasoneofthebestprocessesinmicrofluidicfabrication.
Ihavetriedtorepeatthisprocessinmylaboratory,butIhaveaproblemforpeeling
offPDMSfrommastermould.Wouldyoupleasemoreexplainformeaboutthisprocess?The
materialofPetridishthatyouusedisglassorplastic?AndwhenIpourPDMSonthe
master,duringthecuringprocessPDMSadheretothePetridisheitherglassandplastic,
inadditiontheadhesiveproblemrelatedPDMSandmastermould.
Iwanttoexplainmoreaboutmyproblem.InfactIusedglassPetridishandputthe
mastermouldinitandwhenIpourPDMSonthemastermold,itwillbespreadandsomeof
thePDMSpenetrateunderthemastermouldandcausestheadhesionbetweenglasswallof
thePetriDishandmastermouldandasaresultthemastermouldbreakwhenIwantto
peeloffPDMSandpickupmouldmaster.
IntheotherhandalthoughIusesilanizationprocessforpeelingoffthe
PDMSfrommastermould,itisnoteffective.Ithinktheseproblemsarerelatedtogether.
WouldyoupleaseexplainformehowcanIsolvethem?
Iwouldappreciateyouforhelpingme.

Themastermoldwasfixedinapolystyrenepetridishwithcleanroomadhesivetapetapingtheedge.AsmallamountofliquidPDMSdŒspenetrateunderthewafer,butwecutoutthePDMSdevicewithasurgicalrazorblade.IfyouwanttoreleasePDMSfromthewholewafer,youneedtocutalongtheedgeofthewaferfirstusingarazorblade.

Hello,
Iamcurrentlyabouttostartaprojectinvestigatingglycoproteins-E-selectininovariancancer,andwillneedtouseamicrofluidicdevice.Thisdeviceisbeingfabricatedbythemechanicalengineeringdepartmentinmyuniversity.
However,theyhaveminimalexperienceinthisarea,andhaveaskedmetoresearchwhatthemicrofluidicchannelwouldbecoatedwithforourpurposes.
Iwouldeventuallyneedtobeabletobothimmobiliseproteinsonthewallsofthechannelandmakecellsadhereinamonolayerinthechannel.Iamunabletofindmuchclearinformationwhetherasilanecoatingwillfulfilboththesefunctions,andamlookingformoreinformation.
Couldyoupleasetellmeaboutwhatsortofcoatingwillberequiredinordertobeabletoachieveboththeseaims?
