ExonucleaseIII(E.coli)isa3'->5'exonucleasewhichactsbydigestingonestrandofadsDNAduplexatatimeordigestingtheRNAstrandofanRNA-DNAheteroduplex(1,2).ExonucleaseIII(E.coli)breaksphosphodiesterbondsonthe5'-sideofAPsitesinbothdsDNAandssDNA(3).Itremoves3'-terminalgroupsondsDNA(3),increasesMutYturnover(4),andefficientlydegrades3'-recessedbutnot3'protrudingDNAends(creating5'-overhangs)(5).ExonucleaseIII(E.coli)removesalimitednumberofnucleotidesperbindingevent,resultingincoordinatedprogressivedeletionswithinthepopulationofDNAmolecules(1).
