The B.megaterium expressionsystemprovidesaversatileandeasy-to-handletoolforstableandhigh-yieldproteinproduction,bothsmallandlargescale.

B.megaterium hasproventobeanexcellentalternativehostto E.coli forheterologousgeneexpression.Unlikeotherbacillistrains,proteolyticdegradationbyalkalineproteasesisavoided.Inaddition,therearenoendotoxinsfoundinthecellwall.

HighProteinYield
Asaresult,proteinyieldsareexceptionallyhighevenifinexpensivesubstratesareused.Forexample,theproteinsmutarotase(Mro)andglucosedehydrogenase(Gdh)havebeenaccumulatedupto20%and30%ofthetotalsolubleprotein,respectively.Usingthetightlyregulatedxyloseoperon,thegeneswereinduced130-to350-foldwithoutproteolysis.

ForsecretionvectorsweofferprotoplastsofstrainMS941.BothstrainsaremutantsofDSM319,whereWH320isachemicallymutant,whileMS941hasadefineddeletioninthegeneofthemajorextracellularproteaseNprM.
