MAVS (Mitochondrial antiviral-signaling protein), also known as virus induced signaling adaptor (VISA), IFN-β promoter stimulator I (IPS-1), or caspase activation recruitment domain adaptor inducing IFN-β (CARDIF), is a protein present in the outer membrane of mitochondria and peroxisomes. This protein plays a key role in innate immunity to viral infections. MAVS functions in RNA-sensing pathways, integrating signals from activated sensors, like RIG-1 and MDA5, upon viral infection and ultimately leading to proinflammatory cytokine production and programmed cell death of the infected cells. Recently, these pathways have been identified as targets for immune therapy in cancer, as it has been shown that activation of these pathways can directly induce programmed cell death in the cancer microenvironment.
Currently, few MAVS antibodies have been tested for their specificity. However, the recently launched Invitrogen MAVS Recombinant Rabbit Monoclonal Antibody (Cat. No. 703153) is CRISPR-verified to ensure specificity and has been tested in western blotting.
MAVS rabbit recombinant monoclonal antibody shows human reactivity and is knock-out verified. (A) Western blot analysis was performed using membrane-enriched extracts (30 µg lysate) of HeLa (lane 1), THP-1 (lane 2), A549 (lane 3), Hep G2 (lane 4), and NIH/3T3 (lane 5) cells. A ~60 kDa band corresponding to MAVS was observed only in the human cell lines tested. In Hep G2 cells, the MAVS antibody also recognizes a smaller isoform, evident by a smaller band at ~55 kDa. Detection was performed using the Invitrogen ECL Chemiluminescent Substrate Reagent Kit (Cat. No. WP20005). (B) Antibody specificity was demonstrated by CRISPR-Cas9-mediated knockout (KO) of the target protein in 293T cells. A loss of signal was observed for the target protein in a MAVS KO cell line compared to the Cas9 control cell line.
