Genomemappingandmolecularbreedingoftomato.--2007.pdf(1.5MB,下载次数:0,售价:金钱10)摘要:Thecultivatedtomato,Lycopersiconesculentum,isthesecondmostconsumedvegetableworldwideandawell-studiedcropspeciesintermsofgenetics,genomics,andbreeding.Itisoneoftheearliestcropplantsforwhichageneticlinkagemapwasconstructed,andcurrentlythereareseveralmolecularmapsbasedoncrossesbetweenthecultivatedandvariouswildspeciesoftomato.Thehigh-densitymolecularmap,developedbasedonanL.esculentum×L.pennelliicross,includesmorethan2200Markerswithanaveragemarkerdistanceoflessthan1cMandanaverageof750kbppercM.DifferenttypesofmolecularmarkerssuchasRFLPs,AFLPs,SSRs,CAPS,RGAs,ESTs,andCOSshavebeendevelopedandmappedontothe12tomatochromosomes.MarkershavebeenusedextensivelyforidentificationandmappingofgenesandQTLsformanyBIOLOGicallyandagriculturallyimportanttraitsandoccasionallyforgermplasmscreening,fingerprinting,andmarker-assistedbreeding.TheutilityofMASintomatobreedinghasbeenrestrictedlargelyduetolimitedmarkerpolymorphismwithinthecultivatedspeciesandeconomicalreasons.Also,whenused,MAShasbeenemployedmainlyforimprovingsimply-inheritedtraitsandnotmuchforimprovingcomplextraits.ThelatterhasbeenduetounavailABIlityofreliablePCR-basedmarkersandproblemswithlinkagedrag.Effortsarebeingmadetodevelophigh-throughputmarkerswithgreaterresolution,includingSNPs.TheexpandingtomatoESTdatabase,whichcurrentlyincludes∼214000sequences,thenewmicroarrayDNAchips,andtheongoingsequencingprojectareexpectedtoaiddevelopmentofmorepracticalmarkers.SeveralBAClibrarieshavebeendevelopedthatfacilitatemap-basedcloningofgenesandQTLs.SequencingoftheeuchromaticportionsofthetomatogenomeispavingthewayforcomparativeandfunctionalanalysisofimportantgenesandQTLs.
