 【介绍】
抵抗素(Resistin)，是RSTN基因编码的产物，是一种肽激素，富含半胱氨酸的分泌蛋白，属于RELM家族，也称之为ADSF（脂肪组织特异性的分泌因子）和FIZZ3（存在于炎症发生区）。人的抵抗素含有一段108个氨基酸的前肽，在分泌前被剪切。在外周血中存在的抵抗素是有两段92个氨基酸多肽组成的二聚体，通过Cys26进行二硫键的连接。抵抗素可能是肥胖和胰岛素抵抗之间的重要链接。小鼠的抵抗素，明确由脂肪细胞产生和分泌，作用于骨骼肌细胞、干细胞和脂肪细胞，降低其对胰岛素的敏感性。Steppan et al.研究表明抑制胰岛素刺激糖摄取的能力， 同时在肥胖小鼠血中抵抗素水平升高，而在进行禁食或者使用抗糖尿病药物后下降。在另一个方面，Way et al.研究发现肥胖严重抑制抵抗素的表达，而在严重糖尿病人中的表达增加。其它研究显示小鼠在脂肪细胞分化过程中抵抗素增加，但是抵抗素好像也抑制脂肪形成相反，人的脂肪生成分化则可能与抵抗素基因表达下调有关。抵抗素 (Resistin)是脂肪细胞合成释放的活性多肽，可能参与胰岛素抵抗和糖耐受的调节。所以，推测这种分子可能将肥胖症与胰岛素抵抗连接起来。近期研究结果显示，抵抗素 (Resistin)还是一种重要的脂肪组织旁分泌调质，参与内血管内皮细胞功能稳态的调节，推测抵抗素(Resistin)在二型糖尿病心血管病变发生发展过程中发挥重要病理生理学作用。




 
Principle of the Assay
The AssayMax Human Resistin ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for detection of human resistin in plasma, serum, urine, saliva, and cell culture samples. This assay employs a quantitative sandwich enzyme immunoassay technique, which measures resistin in less than 5 hours. A murine monoclonal antibody specific for resistin has been pre-coated onto a microplate. Resistin standards and samples is sandwiched by the immobilized antibody and a biotinylated polyclonal antibody specific for resistin, which is recognized by a streptavidin-peroxidase conjugate. All unbound material is then washed away and a peroxidase enzyme substrate is added. The color development is stopped and the intensity of the color is measured.
