Abstract
Purpose:Exposuretoradiationprovokescellularresponses,whicharelikelyregulatedbygeneexpressionnetworks.MicroRNAsaresmallnon-codingRNAs,whichregulategeneexpressionbypromotingmRNAdegradationorinhibitingproteintranslation.TheexpressionpatternsofbothmRNAandmiRNAduringtheradiation-inducedlunginjury(RILI)remainlesscharacterizedandtheroleofmi-RNAsintheregulationofthisprocesshasnotbeenstudied.ThepresentstudysoughttoevaluatemiRNAandmRNAexpressionprofilesintheratlungafterirradiation.
Methodsandmaterials:MaleWistarratsweresubjectedtosingledoseirradiationwith20Gyusing6MVx-raystotherightlung.(Adoserateof5Gy/minwasapplied).Ratsweresacrificedat3,12and26weeksafterirradiation,andmorphologicalchangesinthelungwereexaminedbyhaematoxylinandeosin.ThemiRNAandmRNAexpressionprofileswereevaluatedbymicroarraysandfollowedbyquant-itativeRT-PCRanalysis.
Results
A
CDNAmicroarrayanalysisfound2183transcriptsbeingup-regulatedand2917transcriptsdown-regulated(P<=0.05,>=2.0foldchange)inthelungtissuesafterirradiation.Likewise,amiRNAsmicroarrayanalysisindicated15miRNAspeciesbeingup-regulatedand8down-regulated(P<= 0.05). Subsequent bioinformatics anal -yses of the differentially expressed mRNA and miRNAs revealed that alterations in mRNA expression following irradiation were negatively correlated with miRNAs expression.
Conclusions
OurresultsprovideevidenceindicatingthatirradiationinducesalterationsofmRNAandmiRNAexpressioninratlungandthatthereisanegativecorrelationofmRNAandmiRNAexpressionlevelsafterirradiation.Thesefindingssignificantlyadvanceourunderstandingoftheregulatorymechanismsunderlyingthepathophysiologyofradiation-inducedlunginjury.Insummary,RILIdoesnotdevelopgraduallyinalinearprocess.Infact,differentcelltypesinteractviacytokinesinaverycomplexnetwork.Fur
Thermore,thisstudysuggeststhatmicroRNAsmayserveanimportantroleinthepathogenesisofRILIandthatunderstandingtheirroleinRILImayhaveasignificanteffectonpatientmanagementanddiagnosisinthefuture.
