| ScientificBackground: | Sincethediscoveryofsomatostatin-14(SOM-14)inovinehypothalamus,severalotherformsofsomatostatin-likeimmunoreactantsofdifferentmolecularweighthavebeenreported,includingsomatostatin-28whichexpressesthefullSOM-14sequenceatitsC-terminus(SOM-28[15-28]).InmammalsbothmolecularformsareencodedattheC-terminusofthesomatostatinprecursormolecule.Hundredsofimmunolocalisationstudieshavebeenpublisheddemonstratingtheexistenceofsomatostatin-likemoleculesinneurons,inpancreaticisletDcellsandinendocrinecellsdistributedthroughoutthemammaliangut,aswellasinrareendocrinetumours(so-calledsomatostatinomas).Themolecularformsappeartobeco-packagedindense-coredsecretorygranules,frequentlywithothertransmittersubstances.Ithasbeenshownthatthemajorityofsomatostatin-containingaxonsintheratspinalcordexpressvesiculargluatamatetransporterprotein-2(VGLUT2). |

Fixatives:Fixationbytranscardialperfusionisbest.Foroptimaltissuepreservationandmaintenanceofantigenicity,use4%bufferedparaformaldehyde(4gparaformaldehydeheatedto70°Cin100ml0.1Msodiumphosphatebuffer,pH7.2,allowedtocool).Ashortpre-wash(50-100ml)toremovebloodisadvised.Followingdissectionsmallpiecesoftissueshouldbepost-fixedfor30-60mininthesamefixativeatroomtemperature.Glutaraldehyde(upto0.2%)canbeaddedifrequired.Tissuesunsuitableforperfusionshouldbefixedbyimmersionintheabovefixative,butcareshouldbetakentoavoidover-fixation(smallpiecesoftissuedonotrequiremorethan60miningeneral).
