PODS®Technology

PODS®proteinsaremadeusinganinsectcellexpressionsysteminwhichtheactiveproteinisco-expressedalongsidepolyhedrincarrierprotein.Polyhedrinformsmicrocrystalswithininsectcellswhichspecificallycapturetheactiveproteintoformaco-crystalcomplex.Theactiveproteiniscapturedinitsnascent,nativelyfoldedformwithlimitedscopeforproteolyticdegradation.Consequently,excellentlevelsofbioactivityareobserved.ThePODS®co-crystalsprovideasustainedreleasemechanismandcanbeusedtofunctionalizesurfaces.Forfurtherdetails,pleaserefertothePODS®Technologypage.

ProductDescription

Theproductcontainsthepolyhedrinproteinco-crystalizedwithHumanBMP-4.BoneMorphogenicProtein4(BMP-4)isamemberoftheBoneMorphogeneticProtein(BMP)family.Theseproteinsaresynthesizedaslargeprecursormoleculeswhicharecleavedbyproteolyticenzymes.BMPproteinsstimulatetheproductionofbonematrixproteinsandosteoclastsproliferation,andcanbefoundontissuesrelatedwithboneorcartilagegrowth.

UsageRecommendation

PODS®co-crystalsprovideadepotofproteinswhicharesteadilysecreted.Ithasbeenestimatedthatthebiologicalactivityof50millionPODS®co-crystalsgeneratesthesamepeakdoseas3.3μgofstandardrecombinantprotein.However,at5daysfollowingthestartofseedingthePODS®co-crystals,therearemorethan50%ofthesepeaklevelsstillpresentintheculturesystem.Ultimately,theamountofPODS®co-crystalsthatisoptimalforaparticularexperimentshouldbedeterminedempirically.Basedonpreviousdata,wesuggestusing50millionPODS®co-crystalsinplaceof3.3μgofstandardgrowthfactorasastartingpoint."

Tocontrolforcross-reactivitywithcellsorasanegativecontrol,werecommendusingPODS®growthfactorsalongsidePODS®Emptycrystals,asthelatterdonotcontainorreleasecargoprotein.
