Biotin,alsoknownasvitaminH,isofgreatimportanceforthebiochemistryofthehumanorganism.Asaprostheticgroupofmitochondrialenzymes(carboylases),biotinplaysacentralroleasaCO2-carrierinimportantmetabolicreactionssuchasgluconeogenesis,synthesisoffattyacidsandmetabolismofaminoacids.FurThermore,biotininfluencesthegrowthandmaintenanceofbloodcells,sebaceousglands,skin,hairandnails.Nexttothefreeformofbiotin,thebiotinlinkedtolysin,alsoknownasbiocytine,canalsobeutilizedasavitaminsourcebythebody,aftercleavagefromtheproteinbytheenzymebiotinidase.
Innature,biotinisverycommon.Itcanbefoundinbacteriaandmushrooms,aswellasinhigherplantlifeandanimaltissue-especiallyinliverandkidney.However,thebiotinavailABIlityinsomefoodisveryslim.Amountsworthmentioningcanonlybefoundinyeast,soyabeans,nuts,cauliflower,lentils,oats,wheatgermandineggyolkwhereasfruits,milkproductsandmostvegetablesonlycontainsmallamounts.Solely,theamountofbiotinisnotthedecisivefactor,butratherthebioavailability.Thebioavailabilityvariesverystronglyanddependsonthekindoffoodandalsoontheextenttowhichbiotinisprotein-bound.Whereas,forexample,inplants,biotinisavailableinthefreeform,foodcomingfromanimalscontainsmostlyprotein-boundbiotin.Onlyafterproteolyticreductioninthesmallintestinefollowedbyacleavagewithintestine(pancreas)-biotinidaseisthebiotinavailableinthefreeabsorbableform.
Biotinwhichissynthesizedendogenouslybyfloraoftheintestineisnotreabsorbedinthecolon,butisstoredasprotein-boundbiotininthebacteriaoftheintestineandisthusnotavailabletocoverthebiotinrequirementoftheorganism.Thetreatmentoffoodalsocauseslossesinbiotin.Wheat,asawholegrain,contains4timesasmuchbiotinasall-purposeflour(Type405).Thealimentaryutilizationofbiotinisestimatedat50%,sothatabiotindeficiencycaneasilyresult.Malnutritionaswellasaninheriteddisorderinbiotinmetabolism(singularormultipledeficiencyincarboxylase,biotinidasedeficiency)canleadtoabiotindeficiency.Furthermore,circumstancesoflife,whereanincreasedbiotindemandexists(pregnancy,nursing,athleticactivities,pathologicalconditions)maycauseabiotinmalnutrition.Avarietyofdisordersonhair,skinandnailsarethemedicallyrelevantconsequencesofabiotindeficiency.Thesymptomsrangefrombrittle,splinteredfingernailstodifferentformsofalopeciatoscalyerythematousandseborrheicdermatitis.Animalshavealsobeenobservedwithsimilarillnesses,rangingfromlargerskindetachments,epidermalcrustdevelopment,aswellasahyper-andparakeratosicalchangeinmucousmembrane.
