Bisphosphonatesbindstronglytoboneandreduceboneresorptionthrougheffectsonbothosteoclastsandosteoblasts.Clodronateisanon-nitrogenousbisphosphonatethathasbeenusedinthetreatmentofosteoporosisandothermetabolicbonediseases.Itbindstohydroxyapatitecrystalswithanaffinityconstantof0.72µM,inhibitingtheirgrowth(IC50=1.34µM)andboneresorption.Clondronateismetabolizedintracellularlytoaβ-γ-methyleneanalogofATPthatiscytotoxictobothosteoclastsandmacrophages,whichbelongtothesamecelllineageasosteoclasts.Liposome-encapsulatedbisphosphonatessuchasclodronatehavebeenusedinbiologicalresearchtoselectivelydepletemacrophage-likecellsandcancercells.
