Description
Cedarlane’santi-Mac-2monoclonalantibodyspecificallybindsthemouseMac-2antigen(alsocrossreactswithhuman).Theantibodyrecognizesa32,000daltonsurfaceantigenfoundonasubpopulationofmousemacrophages.Itreactswithperitonealexudatemacrophageswheretheexudateisprovokedbythioglycollate,proteasepeptone(20%),macrophagesoflymphoidandnon-lymphoidtissues,interdigitatingdendriticcellsandLangerhanscells.Mac-2isalsoexpressedinthecytoplasmonnon-elicitedresidentmacrophages;5%arestronglyreactive,theremaining95%showmuchweakerstaining.
TheantibodydoesnotreactwithperitonealexudatemacrophageswheretheexudateisprovokedbyListeriamonocytogenes,lipopolysaccharideorconcanavalinA.Italsodoesnotreactwithperitonealmacrophages,splenicmacrophages,granulocytes,thymocytes,peripherallymphnodecellsandwith99%ofbonemarrowcells. 
CL8942antibodycanbeusedforindirectimmunofluorescencestaining,includingflowcytometricanalysisoflivecells.Theadditionofpropidiumiodideisoptional;itsuseeliminatesstainingartifactscausedbydeadcells(1).Thisantibodyisalsosuitableforfrozensections(4,5)andparaffinsections(7)(recommendedstartingdilutionsof1:1000-1:2000);ELISA(6);andWesternBlot.______________________________________________________________________________________________________
