ForprintingwithLifeink®200,wehighlyrecommendusingtheFRESHprintingtechniqueandprintingat2-8°C.Threedimensional(3D)gelsallowforthestudyoftheeffectsofthemechanicalpropertiesoftheextracellularmatrix(ECM),suchasdensityandrigidity,oncelldevelopment,migration,andmorphology.Unlike2Dsystems,3Denvironmentsallowcellextensionstosimultaneouslyinteractwithintegrinsonallcellsurfaces,resultingintheactivationofspecificsignalingpathways.Gelstiffnessorrigidityalsoaffectscellmigrationdifferentlyin3Dversus2Denvironments.

  1. Placesterilecouplerontheendofthesyringewiththecellsuspension.
  2. Slowlypushplungerinuntilmediaformsaslightexternalmeniscusabovetheendofthecoupleronthesyringe.
  3. Removecapfromthesyringewithcollagenandslowlypushplungerinuntilcollagenformsaslightexternalmeniscusabovetheendofthesyringe.
  4. Couplethesyringewithcellstothesyringewithcollagen.(Ensurethattherearenoairbubblesinthesystem.The“externalmeniscus”onbothsyringeshelpsensurethattherearenoairbubblesintroduced).
  5. Slowlypushplungersbackandforth~40timestoensurethoroughmixing.Endwithallofthematerialinthesyringetobeusedforprinting.

  1. Thecell-ladenbioinkisnowreadyforpositivedisplacementprinters.Allow5-10minutesforthesyringetoequilibratebackto2-8°Cbeforeprintingforbestresults.

Note:Forpneumaticprinters,transferthecollagenintoanappropriatesyringeusingthecoupler.Thenewsyringeshouldhavethesealinserted,buttheplungerremoved.Centrifugethesyringeat2000RPMfor1minuteaftertransferringthecollagentoremoveanyairbubbles.Donotdothecentrifugestepiftherearecellsalreadymixedin.

GeneralPrintingNotes:

  1. Touseasmallervolumeofcollagen,simplytransferthedesiredamountofcollagentoanothersyringe,usingtheprovidedsterilecoupler.Toremovetheairfromthenewsyringe,youcandoeitherofthefollowing:
  2. Centrifugethesyringe(capped)withthecappointinguptocausetheairtoaccumulateatthecap.Evacuatetheair.
  3. Centrifugethesyringe(capped)withthecappointingdown,andthenuseahemostattosqueezethesyringewhilepushingtheplungertoallowtheairtoescape.

Removingairwithahemostatvideo

  1. Completeyourprintwithin1hourofcelladditiontosyringe/bioinkforbestcellviABIlityresults.
  2. WhenprintingwithFRESHgelatinslurry,allowthefinalprintedstructuretoincubateat37°Cfor30to60minutesandthenreplacethegelatinwithmedia.
  3. Avoidbubbles.
