5µgoftotalproteinfromArabidopsisthaliana(1),Hordeumvulgare(2),Zeamays(3) extractedwithAgriseraPEBbuffer(AS08300) wereseparatedon 4-12%NuPAGEBis-Trisgel(Invitrogen)andblotted1htoPVDF.BlotswereblockedwithECLAdvanceblockingreagent for1.5hatroomtemperature(RT)withagitation.Blotwasincubatedintheprimaryantibodyatadilutionof1:10000for1hatRTwithagitation.Theantibodysolutionwasdecantedandtheblotwasrinsedbrieflytwice,thenwashedoncefor15minand3timesfor5mininTBS-TatRTwithagitation.Blotwasincubatedinsecondaryantibody(anti-rabbitIgGhorseradishperoxidaseconjugated,Agrisera,AS09602)dilutedto1:25000in for1hatRTwithagitation.Theblotwaswashedasaboveanddevelopedfor5minwithECLaccordingtothemanufacturersinstructions.Exposuretimewas44seconds.
