30µgoftotalproteinfromArabidopsisthaliana,ecotypeWs-0extractedwithextractionbuffer(100mMTricine,3mMMgSO₄,3mMEDTA,1mMDTT)wereseparatedon12%SDS-PAGEandblotted1htoPVDF(Biorad).Blotswereblockedwithnon-fatdrymilk(Biorad)for1hatroomtemperature(RT)withagitation.Blotwasincubatedintheprimaryantibodyatadilutionof1:250for1hatRTwithagitation.Theantibodysolutionwasdecantedandtheblotwasrinsedbrieflytwice,thenwashedoncefor15minand3timesfor10mininTBS-TatRTwithagitation.Blotwasincubatedinsecondaryantibody(goatanti-henIgY(H&L),horseradishperoxidaseconjugated)dilutedto1:0000innon-fatdrymilk(Biorad)for1hatRTwithagitation.Theblotwaswashedasaboveanddevelopedfor1minwithECLPlusWesternBlottingDetectionSystem(GEHealthcare)accordingtothemanufacturersinstructions.Exposuretimewas30seconds.
