10µgoftotalprotein from Arabidopsisthaliana (1), Hordeumvulgare (2), Zeamays (3) totalproteinfromallthesampleswereextractedwithProteinExtractionBufferPEB(AS08300).Samplesweredilutedwith1Xsamplebuffer(NuPAGELDSsamplebuffer(Invitrogen)supplementedwith50mMDTTandheatat70Cfor5minandkeeponicebeforeloADIng.ProteinsampleswereseparatedonNuPAGE4-12%Tris-Bisgel(Invitrogen)LDS-PAGEandblottedfor1hto1.5honPVDFusingtanktransfer.Blotswereblockedimmediatelyfollowingtransferin2%blockingreagent(GERPN2125;Healthcare)or5%non-fatmilkdissolvedin20mMTris,137mMsodiumchloridepH7.6with0.1%(v/v)Tween-20(TBS-T)for1hatroomtemperaturewithagitation.Blotswereincubatedintheprimaryantibodyatadilutionof1:5000(inblockingreagent)for1hatroomtemperaturewithagitation.Theantibodysolutionwasdecantedandtheblotwasrinsedbrieflytwice,andthenwashed1x15minand3x5minwithTBS-Tatroomtemperaturewithagitation.Blotswereincubatedinsecondaryantibody(anti-rabbitIgGhorseradishperoxidaseconjugated,recommendedsecondaryantibody AS09602)dilutedto1:25000inblockingreagentfor1hatroomtemperaturewithagitation.Theblotswerewashedasabove.Theblotwasdevelopedfor5minwithTMA-6(lumigen)detectionreagentaccordingthemanufacturersinstructions.ImagesoftheblotswereobtainedusingaCCDimager(FluorSMax,Bio-Rad)andQuantityOnesoftware(Bio-Rad).

WT-Col-0 Arabidopsisthaliana leaveswerefrozeninliquidN2andsolubleproteinswereextractedinabuffercontaining50mMHEPES,5mMNaCland10mMMgCl2.10µgand17ugofnativeproteincomplexeswereseparatedbynon-reducingbluenativePAGE(5-12,5%)andthenfurtherseparatedinaseconddimensionbyreducingSDS-PAGE(15%with6Murea)andblotted1htoaPVDFmembraneusingHöefersemi-dryblotter.Blotswereblockedwith2%milkinTTBSfor3hatroomtemperature(RT)withagitation.Blotwasincubatedintheprimaryantibodyatadilutionof1:2000overnightin+4°C.Theantibodysolutionwasdecantedandtheblotwaswashed3x5minwithTTBSatRTwithagitation.Blotwasincubatedinsecondaryantibody(anti-rabbitIgGhorseradishperoxidaseconjugated,fromAgrisera AS09602)dilutedto1:25000in1%milk/TTBSfor2hatRTwithagitation.Theblotwaswashed3x5minutesinTTBSand1x5mininTBSatRTwithagigationanddevelopedfor5minwithECLaccordingtothemanufacturer"sinstructions.Exposuretimewas30seconds.

Totalproteins(40µg)from Chlamydomonasreinhardtii strainCC-124 (1), Chlamydomonasreinhardtii strainCC43-48(STA6) (2) andPolytomella sp.Pringsheim198.80 (3) wereseparatedona5-12%acrylamide/6MureaSDS-PAGEandblottedontonitrocellulosemembrane.BlotswereblockedinT-TBS(0.1%TweeninTrisBufferSaline)containing5%lowfatmilk,1hatroomtemperature(RT)withagitation.Blotwasincubatedintheprimaryantibodyatadilutionof1:2500for1hatRTwithagitation.Theantibodysolutionwasdecantedandtheblotwaswashedtwicefor15mininT-TBS,RTwithagitation.Blotwasincubatedinsecondaryantibodydilutedto1:25000(goatanti-rabbitIgG,horseradishperoxidaseconjugated,Agrisera, AS09602),1hatRTwithagitation.Theblotwaswashedasaboveanddevelopedusingthehomemadeenhancedchemiluminescencesystem(Durrant,Nature346:297,1990).ImagesoftheblotswereobtainedusingaCCDimager(ImageQuantLAS4000).
