ImmunoglobulinA(IgA)isthemainimmunoglobulininmucoussecretionsandthesecondmostabundantimmunoglobulininserum.ItdefendsagainstinhaledandingestedpathogensbybindinganFcreceptoronmyeloidleukocytes.Theequivalentmousereceptorhasnotyetbeenidentified.HumanIgAhastwodistinctsubclassesandismonomericinserumwhilemouseIgAdoesnothavesubclassesandisdimericinserum.ThesensitivequantitativemeasurementoftotalmouseIgAantigeninserum,plasma,hybridomacellsupernatants,ascitesorotherBIOLOGicalfluidsamplesiseasilyperformedwiththis96wellstripformatELISAkit.Thekitdoesnotcrossreactsignificantlywithrat,pig,sheep,dog,rabbit,cynomonkey,rhesusmonkey,orhumanIgA.TheconcentrationofIgAwasfoundtobe0.4mg/mlinNIHSwissmouseserumand0.7mg/mlinBALB/Cmouseserum.TheassaymeasuresmouseIgAinthe0.1-100ng/mlrange.SamplesgivingmouseIgAlevelsabove100ng/mlshouldbedilutedinblockingbufferbeforeuse.A1:200,000dilutionforserumissuggestedforbestresults.MouseIgAwillbindtothecaptureantibodycoatedonthemicrotiterplate.Afterappropriatewashingsteps,peroxidaselabeledanti-mouseIgAprimaryantibodybindstothecapturedprotein.ExcessantibodyiswashedawayandTMBsubstrateisusedforcolordevelopmentat450nm.ColordevelopmentisproportionaltotheconcentrationofmouseIgAinthesamples.AstandardcalibrationcurveispreparedinblockingbufferusingdilutionsofpurifiedmouseIgAandismeasuredalongwiththetestsamples.AllreagentsandstandardsareprovidedintheseELISAkits.
