N-[6-(Biotinamido)hexyl]-3’-(2’-pyridyldithio)-propionamide(Biotin-HPDP)isasulfhydrylreactivebiotinylationreagentwithalongchain,cleavablespacerarm.Biotin-HPDPbiotinylationoccursbetweenpH6-9throughitspyridyldithiolgroupreactingwithsulfhydrylstoformastabledisulfidebond.The29.2angstromspacerarmpreventssterichindranceandmayalsobecleavedatthedisulfidebondusingTCEP-HCl,DTTorß-Mercaptoethanol.Biotin-HPDPmustbedissolvedinanorganicsolvent,suchasDMSOorDMF,thenaddedtoanaqueousbiotinylationreaction.TheresultingbiotinylatedproteincanthenbedetectedwhenusedinconjunctionwithstreptavidinorImmobilizedStreptavidin.
