Antibodiesandotherconjugates
Theguaranteedshelflifefromdateofreceiptforantibodiesandconjugatesislistedontheproductinformationsheet.Antibodiesandotherconjugatesoftenarefunctionalforsignificantlylongerthantheguaranteedshelflife.Ifyouhaveanolderconjugateinstoragethatyouwishtouse,werecommendperformingasmallscalepositivecontrolexperimenttoconfirmthattheproductstillworksforyourapplicationbeforeprocessingalargenumberofsamplesorprecioussamples.
Forlyophilizedantibodies,werecommendreconstitutingtheantibodywithglycerolandantimicrobialpreservativelikesodiumazideforthelongestshelflife(notethatsodiumazideisnotcompatiblewithHRP-conjugates).
Chemicals,dyes,andgelstains
Biotiumguaranteesthestabilityofchemicals,dyes,andgelstainsforatleastayearfromthedateyoureceivetheproduct.However,themajorityoftheseproductsarehighlystableformanyyears,aslongastheyarestoredasrecommended.Storageconditionscanbefoundontheproductinformationsheetorproductsafetyanddatasheet,materialsafetydatasheet,andontheproductlabel.Fluorescentcompoundsshouldbeprotectedfromlightforlongtermstorage.
IfyouhaveaBiotiumcompoundthathasbeeninstorageforlongerthanoneyearthatyouwishtouse,werecommendperformingasmallscalepositivecontrolexperimenttoconfirmthatthecompoundstillworksforyourapplicationbeforeprocessingalargenumberofsamplesorprecioussamples.

Estercompoundsincludethefollowing:

Esterdyesarestableinsolidformaslongastheyareprotectedfromlightandmoisture.Estersarenotstableinaqueoussolution.ConcentratedstocksolutionsshouldbepreparedinanhydrousDMSO(seeBiotiumcatalogno.90082).StocksolutionsinanhydrousDMSOcanbestoreddesiccatedat-20°Cforonemonthorlonger.Estersshouldbedilutedinaqueoussolutionimmediatelybeforeuse.Succinimidylesters(SE)shouldbedissolvedinasolutionthatisfreeofamine-containingcompoundslikeTris,glycine,orprotein,whichwillreactwiththeSEfunctionalgroup.AMestersanddiacetatecompoundsshouldbedissolvedinasolutionthatisfreeofserum,becauseserumcouldcontainesterasesthatwouldhydrolyzethecompound.
AnoteonCF™dyesuccinimidylesterstability
Succinimidylestersaregenerallysusceptibletohydrolysis,whichcanresultinlowerlabelingefficiency.Heavilysulfonateddyes,suchastheAlexaFluor®dyes,DyLight®dyesandIRDyes®areparticularlyhygroscopic,worseningthehydrolysisproblem.Forexample,thepercentofactiveAlexaFluor®488succinimidylester(SE)couldbewellbelow50%bythetimeofapplication(accordingtothemanufacturer’sproductdatasheet).InanumberofAlexaFluor®SEreactivedyes,theSEgroupisderivedfromanaromaticcarboxylicacid,whileinallofBiotium’sCF™dyestheSEgroupispreparedfromanaliphaticcarboxylicacid.ThisstructuraldifferencereducesthesusceptibilityofCF™dyeSEreactivegroupstohydrolysis,resultinginrelativelystablereactivedyeswithconsistentlyhigherlabelingefficiencycomparedtootherSEderivativesofotherfluorescentdyes.
Maleimides,MTSandthiosulfatedyes
Likethesuccinimidylesterdyes,thesedyesarealsosusceptibletohydrolysis,althoughgenerallytoamuchlowerdegree.Thus,forlongtermstorage,anhydrousDMSOisrecommendedformakingstocksolutions.
Otherreactivedyes
Amines,aminooxy(alsoknownasoxylamine),hydrazide,azide,alkyne,BCN,andtyramidereactivedyes,aswellasdyefreeacids,aregenerallystableinaqueoussolutionwhenstoredat-20°Cfor6-12monthsorlonger,aslongasnocompoundsarepresentthatmayreactwiththedye’sfunctionalgroup.Seetheproductinformationsheetsforspecificreactivedyesmoreinformation.
CoelenterazinesandD-luciferin
Coelenterazinesarestableinsolidformwhenstoredasrecommended;theyarenotstableinaqueoussolution.Concentratedcoelenterazinestocksolutions(typically1-100mg/mL)shouldbepreparedinethanolormethanol;donotuseDMSOorDMFtodissolvecoelenterazines,becausethesesolventswilloxidizethecompounds.Ethanolormethanolstocksofcoelenterazinecanbestoredat-20°Corbelowforsixmonthsorlonger;alcoholstocksmayevaporateduringstorage,sousetightlysealingscrewcapvialsandwrapthevialswithParafilmforlongtermstorage.Propyleneglycolalsocanbeusedasasolventtominimizeevaporation.Ifthesolventevaporates,thecoelenterazinewillstillbepresentinthevial,sonotethevolumeinthevialpriortostoragesothatyoucanadjustthesolventvolumetocorrectforevaporationifneeded.Prepareworkingsolutionsinaqueousbuffersimmediatelybeforeuse.Coelenterazinesarestableforuptofivehoursinaqueoussolution.
Aquaphile™coelenterazinesarewatersolubleformulationsofcoelenterazines.Theyarestableinsolidformwhenstoredasrecommended.Aquaphile™coelenterazinesshouldbedissolvedinaqueoussolutionimmediatelybeforeuse.Theyarestableforuptofivehoursinaqueoussolution.
Notethatcoelenterazinesarepredominantlyyellowsolids,butmaycontaindarkredorbrownflecks.Thisdoesnotaffectproductstabilityorperformance.Ifyourcoelenterazineisuniformlybrown,thenitisoxidizedandneedstobereplaced.
D-luciferinisstableinsolidformandasaconcentratedstocksolutionwhenstoredasrecommended;itisnotstableatdiluteworkingconcentrationsinaqueoussolution.PrepareconcentratedD-luciferinstocksolutions(typically1-100mg/mL)inwater,andstoreinaliquotsat-20°Corbelowforsixmonthsorlonger.Prepareworkingsolutionsimmediatelybeforeuse.

Otherfluorescentdyesintransparentvialsshouldstoredinthedarkforlongtermstorage.Itsfinetostoredyevialsuncoveredinawindowlessrefrigeratororfreezerwithanautomaticlightshut-off.Ifdyeswillbestoredinaglass-front(deli-style)refrigerator,walk-inrefrigerator/freezer,oratroomtemperaturewithconstantlightexposure,theyshouldstoredinanon-transparentbox(likeawhitecardboardfreezerbox),inacloseddrawer,inablackplasticbag,orcoveredwithaluminumfoil.
Fluorescentdyesgenerallyarenotsensitivetobrieflightexposurewhiletheyarebeinghandledonthebenchduringanexperiment.Tobeonthesafeside,weusuallylooselycovertuberackswithapieceoffoilifthedyevialsaregoingtobeoutonthebenchformorethan30minutesorso. Butmostdyesarestableenoughthatevenaccidentallyleavingthemonthebenchforadaywillnotaffectperformance.WhenhandlingphotoreactivedyeslikeEMA,PMA,orPMAxx,wetaketheextraprecautionofdimmingthelights.
