TheFab-TACSGravitycolumnisfilledwithaStrep-Tactin®coatedmatrixmadeofcell-gradeagarose.Strep-taggedFabfragments(Fab-Streps)specificallybindtothematrix.Subsequently,wholebloodorotherbloodpreparationspassthroughthecolumn.TargetcellsadheretothematrixbasedontheexclusivebindingoftheFab-Streptothetargetcell.Non-targetcellsareefficientlywashedaway.Inafinalstep,theadditionofbiotincausestheelutionofthetargetcellsandtheFab-Streps.Afterelution,theFab-Strepsself-dissociatefromthecellsurfaces.Thelabel-freeauthentictargetcellsarenowreadyforfurtherdownstreamapplications.
