Humanimmunodeficiencyvirus(HIV)andrelatedvirusesrequirecoreceptorsinadditiontoCD4toinfecttargetcells.SomeGprotein-coupledreceptorsinthechemokinereceptorfamily,includingCCR5,CXCR4,CCR3,CCR2b,andCCR8inthechemokinereceptorfamily,havebeenidentifiedasHIVcoreceptors.CXCR4(alsocalledFusin,LESTR,HUMSTR)isaprincipalcoreceptorforT-celltropicstrainsofHIV-1.CXCR4isalsorequiredfortheinfectionbydual-tropicstrainsofHIV-1andmediatesCD4-independentinfectionbyHIV-2.Theα-chemokineSDF-1istheligandforEXCR4andpreventsinfectionbyT-celltropicHIV-1.CXCR4associateswiththesurfaceCD4-gp120complexbeforeHIVenterstargetcells.Theamino-terminaldomainandthesecondextracellularloopofCXCR4serveasHIVbindingsites.

CXCR4messengerRNAlevelscorrelatewithHIV-1permissivenessindiversehumancelltypes.AntibodiestoCXCR4mayblockHIV-1andHIV-2infectionofhumantargetcells.
