Description

ThethreemammalianisoformsofTGF-β,TGF-β1,β2,andβ3,signalthroughthesamereceptorandelicitsimilarbiologicalresponses.Theyaremultifunctionalcytokinesthatregulatecellproliferation,growth,differentiationandmotility,aswellassynthesisanddepositionoftheextracellularmatrix.Theyareinvolvedinvariousphysiologicalprocesses,includingembryogenesis,tissueremodelingandwoundhealing.Theyaresecretedpredominantlyaslatentcomplexes,whicharestoredatthecellsurfaceandintheextracellularmatrix.ThereleaseofthebiologicallyactiveTGF-βisoformfromalatentcomplexinvolvesproteolyticprocessingofthecomplexand/orinductionofconformationalchangesbyproteinssuchasthrombospondin-1. TGF-β1isthemostabundantisoformsecretedbyalmosteverycelltype.Itwasoriginallyidentifiedforitsabilitytoinducephenotypictransformationoffibroblasts,andrecentlyithasbeenimplicatedintheformationofskintumors.RecombinantHumanTGF-β1isa25.0 kDaproteincomposedoftwoidentical112 aminoacidpolypeptidechainslinkedbyasingledisulfide bond.
