Description
Persephinisadisulfide-linked,homodimeric,neurotrophicfactorstructurallyrelatedtoGDNF,artemin,andneurturin.Theseproteinsbelongtothecysteineknotfamilyofgrowthfactorsthatassumestabledimericstructures.Persephinsignalsthroughamulticomponentreceptorsystem,composedofRETandoneoffourGFRα(α1-α4)receptors.TheGFRα4wasfirstidentifiedinchicken,andwaslatershowntobethepreferentialbindingsubunitforpersephin.Persephinpromotesthesurvivalofventralmidbraindopaminergicneuronsandmotorneuronsaftersciaticnerveoxotomy,and,likeGDNF,promotesuretericbudbranching.However,incontrasttoGDNFandneurturin,persephindoesnotsupportthesurvivalofperipheralneurons.RecombinantMurinePersephinisadisulfide-linkedhomodimer,composedoftwo10.3 kDapolypeptidechains(192 totalaminoacidresidues).Eachchaincontainssevenconservedcysteineresidues,oneofwhich(Cys63)isusedforinter-chaindisulfidebridging,andtheothersareinvolvedintheintramolecularringformationknownasthecysteineknotconfiguration.
