Background:

ThereactivityoftheantiserumisdirectedtotheFcandFabsubunitsoftheIgGmolecule.ItincludessomereactivitywithotherimmunoglobulinsviathecommonFabportion.Itdoesnotreactwithanynon-Igproteininbovineserum,astestedbyimmunoelectrophoresisanddoublerADIalimmunodiffusion.ToidentifyandmeasureIgG,antigenorantibody,atthecellularandsubcellularlevelbyimmunofluorescencestainingofappropriatelytreatedcellandtissuesubstrates,andtodemonstratecirculatingantibodiesinSEROdiagnosticmicroBIOLOGyandautoimmunediseases;toidentifyaspecificantigenorimmunecomplexusingareferenceantibodyofbovineorigininthemiddlelayeroftheindirecttestprocedure.Thisimmunoconjugateisnotpre-diluted.Theoptimumworkingdilutionofeachconjugateshouldbeestablishedbytitrationbeforebeingused.Excesslabelledantibodymustbeavoidedbecauseitmaycausehighunspecificbackgroundstainingandinterferewiththespecificsignal.

CrossReactivity:

Inter-speciescross-reactivityisanormalfeatureofantibodiestoimmunoglobulins,sinceIgofdifferentspeciesfrequentlyshareantigenicdeterminants.Cross-reactivityofthisantiSerumhasnotbeentestedindetail,howevertheconjugatehasbeentestedindoubleradialimmunodiffusion(Ouchterlony)antiHumanandSwineSerumandnoreactionhasbeenobserved.

Thelyophilizedconjugateisshippedatambienttemperatureandmaybestoredat+4°C;prolongedstorageatorbelow-20°C.Priortouse,analiquotisthawedslowlyinthedarkatambienttemperature,spundownagainandusedtoprepareworkingdilutionsbyaddingsterilephosphatebufferedsaline(PBS,pH7.2).Repeatedthawingandfreezingshouldbeavoided.Workingdilutionsshouldbestoredat+4°C,notrefrozen,andpreferablyusedthesameday.Ifaslightprecipitationoccursuponstorage,thisshouldberemovedbycentrifugation.Itwillnotaffecttheperformanceoftheimmunoconjugate.
