GentargetprovidesdifferentsetsofpremadelentivirialparticlesforhumanormouseiPSgenes(seeiPSparticleproductpage).

ThispremadelentiviralparticlesisthesinglevectormethodforiPScellgeneration.FourhumaniPSfactor,c-Myc,Klf4,Oct4andSox2(MKOS)wereclonedunderthesameTetracyclineinducibleCMVpromoter.All4factorswereclonedinframebutseparatedby2Asignalfragment,whichresultintheexpressionoffourindividualfactorvia“ribosomalskipping”mechanism.Inaddition,theparticlescontainsaGFP-blasticidindualselectionmarkerunderRSVpromoter,whichprovidesaconvenience methodtomonitortheiPSfactorexpressionefficiency,andtosortthepositivetransduced cells.

ThefouriPSfactorareexpressedunderanoptionalinducibleCMVpromoter.ThepremadeparticlecanbeusedasconstitutiveexpressionoffouriPSfactors,butoptionally,itcanbeusedasantetracyclineinducibleexpressionofiPSfactors.Tousedasinducibleexpression,therepressprotein(TetR)mustbepresentinadvancetostoptheexpression,theexpressionofiPSfactorsonlyoccurswhentheTetracyclineorDoxwasaddedintothecellculture.PleasetoseemoredetailsabouttheOptionalinduciblelentiviralexpression.
