 Host/IsotypeMouse/IgG2b Clone27B10.4 SpeciesReactivityAllspecies | Anti-phosphotyrosine-HRPantibodyisanHRPlabeledtyrosinephosphorylationspecificantibodythatispartoftheSignal-Seeker™productline.APY03-HRPphosphotyrosineantibodyrecognizesawiderangeoftyrosinephosphorylatedproteinsincellstreatedwithkinaseinhibitors,hydrogenperoxide(H2O2)andsodiumvanadate(Figure1)andhasbeenvalidatedforwesternblotapplications.APY03anti-phosphotyrosineantibody27B10hasbeentestedinwesternblot,immunofluorescence,immunoprecipitation,andELISAwithexcellentspecificityandlowbackgroundwithstrongpotentsignals. APY03isamousemonoclonalantibody27B10thatrecognizesphosphorylatedtyrosineresidues.APY03wasraisedagainstaproprietarymixtureofphosphotyrosinepeptidesconjugatedtoKLH.APY03candetectlessthan10ngofphosphotyrosine-labeledbovineserumalbumin(seeCertificateofAnalysis[COA]).APY03ispurifiedbyproteinGaffinitychromatographyandissuppliedasalyophilizedwhitepowder.EachLotofantibodyisqualitycontrolledtoprovideahighbatchtobatchconsistency.TheLotspecificµgpertubecanbefoundintheLotspecificCOAdocuments.APY03-HRPshowshighspecificitytophosphotyrosineproteinsanddoesnotcross-reactwithphosphoserine/threonineproteinsawesternblotassay(Figures1).

Figure1:WesternBlotdemonstrationofAPY03-HRPphosphotyrosinespecificity A431cellswereeithertreated(+)oruntreated(-)withCalyculinA(CalyA:aserine/threoninephosphataseinhibitor,50nMfor1hour).NIH3T3cellswereeithertreatedoruntreatedwithH2O2-activatedsodiumorthovanadate(Vanadate:aspecifictyrosinephosphataseinhibitor,100µMfor10minutes).10µgofeachlysatewasresolvedinSDS-PAGEandproteinsweretransferredtoPVDFmembrane.APY03-HRP(1:6000)wasusedtodetecttyrosinephosphorylatedproteins.WesternblotwasdevelopedwithSuperSignalWestDurachemiluminescentreagent(ThermoScientific)andexposuretimewas10seconds.AsshowninFig.1,awiderangeoftyrosinephosphorylatedproteinsweredetectedinNIH3T3cellstreatedwithorthovanadatebutnotinCalyculinAtreatedA431cells.
