BACKGROUND

Cortisolisalsoknownashydrocortisonewhenitisusedmedicinally,usuallyasasupplementorforitsanti-inflammatoryproperties.KnowingplasmacortisollevelsisusefulinidentifyingaseriouslackofcortisolusuallyfoundinAddison'sdisease,oraseriousoverproductionofcortisolasinCushing'ssyndrome.Cortisolanalysisisalsofrequentlyusedtoevaluatethestresslevelsofasubject.ThisELISAisdesignedfordetectingcortisolinbiologicalfluidsincludingurine,plasma,andsaliva.

CORTISOLASSAYPRINCIPLE

Thiskitisacompetitiveenzyme-linkedimmunoassay(ELISA)fordetermininglevelsofcortisolinbiologicalsamplesincludingurine,saliva,andplasma.Briefly,cortisolinthesamplesorstandardscompeteswithcortisolconjugatedtohorseradishperoxidase(HRP)forbindingtoapolyclonalantibodyspecificforcortisolcoatedonthemicroplate.TheHRPactivityresultsinbrilliantbluecolordevelopmentwhenthesubstrateisadded,withtheintensityofthecolordevelopmentproportionaltotheamountofcortisol-HRPboundandinverselyproportionaltotheamountofunconjugatedcortisolinthesamplesorstandards.

Whenmeasuringurinesamples,itisrecommendedtonormalizesamplestocreatininetoaccountforvariationsinfluidintakeandexcretion. Creatinineisexcreatedataconstantrateandcanbeusedtonormalizetherateofexcretionofotheranalytes.
