Signosis’stemcellTFactivationprofilingplatearrayisusedformonitoringtheactivationofmultipleTFssimultaneously. Inthetechnology,aseriesofbiotin-labeledprobesaremadebasedontheconsensussequencesofTFDNA-bindingsites. Whentheprobemixincubateswiththecelllysate,individualprobeswillfinditscorrespondingTFandformTF/probecomplexes,whichcanbeeasilyseparatedfromfreeprobesthroughsimplespincolumnpurification. Theboundprobesaredetachedfromthecomplexandanalyzedthroughhybridizationwithaplate;eachwellisspecificallypre-coatedwithcomplementarysequencesoftheprobes.ThecapturedDNAprobeisfurtherdetectedwithstreptavidin-HRP.Luminescenceisreportedasrelativelightunits(RLUs)onamicroplateluminometer.
