TheStandardandSpecialtyseriesofmultiwellchambersandthesinglewellchambersusepunchedfiltersinavarietyofsizes.Theyareavailablewithstandarduntreatedsurfaceorwiththewettingagentpolyvinylpyrrolidone(PVP).
Polycarbonatetrack-etch(PCTE)membranesaretheprimarytypeoffiltersusedinourchambersandinstrumentsystems.Thisisaverythinmembranerangingbetween6-10µminnominalthickness.Thismembranetypeoffersexcellentuniformityinporesizeanddensity,aswellasotherbenefitssuchaslowextractables,lowproteinbinding,andnegligIBLeabsorption.Thesecharacteristicsmakeitideallysuitedforuseinchemotaxisassaysaswellasotherhighpurityscreeningassays.
PVPTreatment–PCTEmembranesareavailablepre-treatedwiththewettingagentpolyvinylpyrrolidone(PVP)whichmakesthesurfacehydrophilic.PVP-treatedmembranesareusedwhenthemediasolutionscannotwetoutthemembranebythemselves.Thisoccursprimarilywhenworkingwithverysmallporesizesorhighviscositymediasolutions.CertaincelltypessuchasneutrophilsdonotadherewelltoPVPtreatedmembranesandtendtodropoffaftermigrating.Whilethisisproblematicwhenquantifyingfiltersonthemembrane,someresearchersusingtheChemoTx®sytemortheMB-series96-wellchambersandquantifyingintheplatefindthistobeanadvantage.Extra-cellularmatrixproteinsdonotadherewelltoPVPtreatedmembraneseither.Ifyourassayrequiressuchcoating,werecommendusingPVP-freemembranes.
