Overview

TheMACSQuant®Tyto™Cartridgehousestheactualheartofthesortingsystem:themicrochip.Thepatentedmicrochiptechnologyenableshigh-speed,fluorescence-basedcellsortingwiththeworld’sfastestmechanicalsortvalve(30,000valveactuationspersecond)inafullyclosedenvironment,providingfullsafetytotheoperator.Inaddition,theTytoCartridgeisasingle-usedisposablethatdiminishesanyriskofsamplecross-contamination.BothpositiveandnegativesortedfractionsremainintheTytoCartridgeaftertheactualsort,allowingfullretrievalofallcellswithinthesample.

TheMACSQuantTytoCartridgedesign
TheTytoCartridgehasthreecompartmentsthatcaneachbeaccessedthroughindividual,asepticLuer-Locksfromabove:theinputchamber,thepositive-sortchamber,andthenegative-sortchamber.
CellssUSPendedinthespeciallyformulatedMACSQuantTytoRunningBufferareloadedintotheinputchamberandarekeptinsuspensionthrougharotatingpropeller.AfterloADIngtheTytoCartridgeintotheinstrument,filteredairisappliedtotheinputchamberthroughthepressureinlet.Drivenbytheairpressure,fluorescentlylabeledcellsflowfromtheinputchamber,throughamicrochannel,intothemicrochipwheretheyareinterrogatedbythreelasers.Fluorescentandscatterlightsignaturesareusedtodeterminewhetheracellispositiveornegative.Toredirectapositivecellintotheappropriatechamber,thesolenoidlocatedontheinstrumentappliesamagneticfieldtothechip,whichtriggersthevalvetoopenupthechanneltothepositivesortchamber.Afterthepositivecellhaspassedthrough,asiliconspringreturnsthevalveintoitsoriginalposition.Unlikeduringconventionaldropletsortingprocedures,thecellsdonotexperiencehighpressure,theyarenotdecompressed,andnochargeisapplied,resultingingreatviABIlityandfunctionalityofcells.
