ODN2006isaB-classODNforactivationofBcells,inductionofIl-6secretionandactivationofNF-kB-signallingpathways.

Backgroundinformation

TLR9isaprominentmemberofthetoll-like-receptor(TLR)familyrecognizingpathogen-associatedmolecularpatterns.TLR9recognizesspecificallyunmethylatedCpGmotifsinbacterialDNAleADIngtoactivationofimmunecells1,2.Theseeffectscanbemimickedbyshortsyntheticaloligodeoxyribonucleotides(ODNs)containingunmethylatedCpGmotifs3.
SeveralclassesofCpGODNhavebeenidentifiedandcanbedistinguishedbytheireffectsoncertaincelltypes.A-classODNscontaining5’and3’G-richstretchesinducehighlevelsoftypeIIFNbutshowlowactivationofBcellproliferation.B-classODNsactivateBcellsandTLR9-dependentNFkBsignallinginrecombinantcelllinesbutshowlowinductionofIFN-α.C-classODNinducehighamountsofIFN-αandactivateBcells.TherecentlydiscoverednewP-ClassODNsshowsimilarbutsuperiorpropertiestoC-classODNs.
