    IKK-γ, also known as NF-kappa-B essential modulator (NEMO) and inhibitor of nuclear factor kappa-B kinase subunit gamma, is the regulatory subunit of the cytoplasmic IκB kinase (IKK) complex. IKK-γ is required for activation of the IKK complex by upstream components of the NF-κB cascade. The IKK complex activates NF-κB which is a ubiquitous transcription factor and an essential mediator of gene expression during activation of immune and inflammatory responses. In the cytoplasm, NFκB is maintained in an inactive form by the inhibitory protein IκB. External stimuli such as tumor necrosis factor or other cytokines results in phosphorylation and degradation of IκB, releasing NF- κB dimers. NF-κB dimers subsequently translocate to the nucleus and activate target genes. IKK-γ interacts preferentially with IKKβ and is required for activation of IKK complex. Recent studies suggest that IKK gamma (NEMO) may be a key molecule acting as an adapter for onco-protein specific signaling to IKKa and IKKb.
