Angiotensinogenistheprecursormolecule,anditisproducedmainlyintheliver,althoughothersiteshavebeenthoughttocontributetolocaleffectsofthemolecule.Productionofthepeptideisincreasedbycorticosteroids,oestrogens,thyroidhomones,andnotably,angiotensinII.Inhumans,thepeptidesequenceforangiotensinogen(485aminoacidslong)whichcontains(immediatlyafterthesignalpeptide)the10aminoacid:Asp-Arg-Val-Tyr-Ile-His-Pro-Phe-His-Leu.Whenbloodpressuredecreasesinthekidneys,theyproduceanenzymecalledrenin.Reninactstocleavethepeptidebondbetweentheleucine(Leu)andthevaline(Val)residues.Thetenaminoacidpeptide(bold)angiotensinIiscreated.||Sequence(linear):Asp-Arg-Val-Tyr-Ile-His-Pro-Phe-His-Leu-Leu-Val-Tyr-Ser||StorageandStABIlity:|Lyophilizedpowdermaybestoredat4°Cforshort-termonly.Stablefor12monthsat-20°C.Reconstitutetonominalvolume(seereconstitutioninstructionsforpeptides)andstoreat-20°C.Formaximumrecoveryofproduct,centrifugetheoriginalvialpriortoremovingthecap.Furtherdilutionscanbemadeinassaybuffer.|||
