IL-10isproducedbymouseTh2cellsfollowingtheirstimulationbylectins.ThemainsourceforBcellderivedIL-10inmiceareLy1BcellsthatexpressCD5(Ly1)andCD11.InhumansIL-10isproducedbyactivatedCD8(+)peripheralbloodTcells,byThelperCD4(+)Tcellclonesafterbothantigen-specificandpolyclonalactivation,byBcelllymphomas,andbymonocytesfollowingcellactivationbybacteriallipopolysaccharidesandmastcells.IL-10isahomodimericproteinwithsubunitshavingalengthof160aminoacids.HumanInterleukin10shows73%aminoacidhomologywithmouseInterleukin-10.ThemurineIL-10receptorhasbeencloned.Thisreceptorisaproteinofapproximately110kDathatbindsmurineIL-10specifically.ThisreceptorisstructurallyrelatedtoreceptorsforIFN.TheCDnomenclatureforthisreceptorisCDw210.IL-10inhibitsthesynthesisofanumberofcytokinessuchasIFN-gamma,IL-2andTNF-betainTh1ThelpersubpopulationsofTcellsbutnotofTh2helpercells.ThisactivityisantagonizedbyIL-4.TheinhibitoryeffectonIFN-gammaproductionisindirectandappearstobetheresultofasuppressionofIL-12synthesisbyaccessorycells.Inthehumansystem,IL-10isproducedby,anddown-regulatesthefunctionof,Th1andTh2cells.
