MesenchymalCellPreparationandUse

HumanBoneMarrow-DerivedMSCsaregeneratedbyculture-expansionofBMMNCsontissueculture-treatedplasticwareinReachbio’sMSCCompleteMedium(catalog#2121).Dependinguponthegrowthcharacteristicsoftheparticularbatch,theyareharvestedatpassagenumber1,2or3(askforCertificateofAnalysisforbatch-specificpassagenumber).Thecellsarevialedandcryopreservedinaserum-freefreezingmediumat0.5millioncells/mL.Uponthawing,theMSCscanbeculturedinMSCCompleteMediumandpassagedanother6-8times.

 

BoneMarrowSampleCollectionandProcessing

Humanbonemarrow(BM)MNCsareobtainedbydensitycentrifugationofBMcells.  BMcellsaredrawnfromtheposterioriliaccrestofnormalhumandonorsusingasyringecontaininganticoagulant. BMcellsareprocessedwithin24-30hoursaftercollection.DonorshavebeenscreenedtobenegativeforHIV,HBV,andHCV.

 

Pre-QualifiedbyFlowCytometry

EachbatchofBM-derivedMSCsispre-qualifiedbyflowcytometrytoensurethecellsexpressthepropercellsurfacephenotypecharacteristicofMSCs.  ThecellsareverifiedtoexpressCD29,CD73,CD90andCD105butlackexpressionofthehematopoieticsurfaceMarkersCD34andCD45. 

 

StABIlityandStorage

BM-derivedMSCsarecryopreservedinCryoStor®CS5,aserum-freefreezingmedium.Cellsarestableforlong-termstorageat-152oCorinthegaseousphaseofliquidnitrogen.Storageat-80oCisnotrecommended. Oncethawed,thecellsmustbeusedimmediatelyandhaveafinitelifespaninculture.
