Peripheralbloodcellsarecollectedfromnormalhumanvolunteersvialeukapheresis(COBE®SpectraMNCcollectionprotocolorequivalent).Themononuclearcell-richleukocytelayer(leukapheresisproduct)iscontinuallyaspiratedbyalowflowratecollectionpumpdirectlyintoacollectionbag.ImmunomagneticcellenrichmentofCD56+cellsisperformedwithin30hoursofcollection.DonorshavebeenscreenedandfoundtobenegativeforastandardpanelofinfectiousagentsthatatminimumincludesHIV,HBVandHCV.Dependingonthespecificlot,otherinfectiousagentsmayalsohavebeentestedfor(CertificateofAnalysisincludeslot-specifictestinginformation).

StABIlityandStorage

PeripheralbloodCD56+NKCellsarecryopreservedinCryoStor®CS5,aserum-freefreezingmedium.Cellsarestableforlong-termstorageat-152oCorinthegaseousphaseofliquidnitrogen.Storageat-80oCisnotrecommended.Oncethawed,thecellsmustbeusedimmediatelyandhaveafinitelifespaninculture.
