SampleCollectionandProcessing

Peripheralbloodcellsarecollectedfromnormalhumanvolunteersvialeukapheresis(COBE®SpectraMNCcollectionprotocolorequivalent).Themononuclearcell-richleukocytelayer(leukapheresisproduct)iscontinuallyaspiratedbyalowflowratecollectionpumpdirectlyintoacollectionbag.ImmunomagneticcellenrichmentofCD14+cellsisperformedwithin30hoursofcollection.DonorshavebeenscreenedandfoundtobenegativeforastandardpanelofinfectiousagentsthatatminimumincludesHIV,HBVandHCV.Dependingonthespecificlot,otherinfectiousagentsmayalsohavebeentestedfor(CertificateofAnalysisincludeslot-specifictestinginformation).

PurityofCD14+Monocytesis>80%(positivelyselected)and80%(negativelyselected)asmeasuredbyflowcytometricanalysis.

 

StABIlityandStorage

PeripheralbloodCD14+monocytesarecryopreservedinCryoStor®CS5,aserum-freefreezingmedium.Cellsarestableforlong-termstorageat-152oCorinthegaseousphaseofliquidnitrogen.Storageat-80oCisnotrecommended.Oncethawed,thecellsmustbeusedimmediatelyandhaveafinitelifespaninculture.
