Description:

Bonemorphogeneticprotein7(BMP-7),alsoknownasosteogenicprotein1(OP1),isawidelyexpressedTGFβsuperfamilymemberwithimportantfunctionsduringembryogenesis,intheadult,andindisease.HumanBMP-7issynthesizedwitha29aminoacid(aa)signalsequence,a263aapropeptide,anda139aagrowthfactordomain.ThegrowthfactordomainofhumanBMP-7shares98%aasequenceidentitywithmouseandratBMP-7.TheBMP-7propeptideiscleavedintracellularlybutoftenremainsassociatedwiththematureC-terminus.Basedoninvivoandinvitrostudies,BMP-7hasthepotentialtobesecretedasadisulfide-linkedmaturehomodimer,orparticularlyasaheteromericcomplexthatconsistsoftwopropeptidesnoncovalentlyassociatedwithamaturedisulfidelinkedhomodimer.ThepresenceofthepropeptidesinBMP-7appearstostabilizethemoleculeandprovideadockingmechanismforextracellularstorageonmoleculessuchasfibrillin1and2.Thepropeptidesthemselvesdonotimpartlatencytothecomplex.BMP-7bindingtotypeIIreceptorsrapidlydisplacestheprodomain:maturemoleculeinteractionandhasnoeffectonactivity.BMP-7playsaroleinavarietyoforgansystems.Itpromotesnewboneformationandnephrondevelopment,inhibitsthebranchingofprostateepithelium,andantagonizesepithelialmesenchymaltransition(EMT).Inpathologicalconditions,BMP-7inhibitstumorgrowthandmetastasis,amelioratesfibroticdamageinnephritis,andpromotesneuroregenerationfollowingbrainischemia.
