H-Yantigenisdefinedasamalehistocompatibilityantigenthatcausesrejectionofmaleskingraftsbyfemalerecipientsofthesameinbredstrainofrodents.Male-specific,orH-Yantigen(s),arealsodetectedbycytotoxicTcellsandantibodies.H-Yantigenappearstobeanintegralpartofthemembraneofmostmalecells.Inaddition,H-YantibodiesdetectasolubleformofH-Ythatissecretedbythetestis.Thegene(Smcy/SMCY)codingforH-YantigendetectedbyTcellshasbeencloned.Itisexpressedubiquitouslyinmalemiceandhumans,andencodesanepitopethattriggersaspecificT-cellresponseinvitro.AdditionalepitopescodedforbydifferentY-chromosomalgenesareprobablyrequiredinvivofortherejectionofmalegraftsbyfemalehosts.ThemolecularnatureofH-Yantigendetectedbyantibodiesonmostmalecellsisnotyetknown.Testis-secreted,solubleH-Yantigen,however,wasfoundtobeidenticaltoMüllerian-inhibitingsubstance(MIS).MIScross-reactswithH-YantibodiesandidenticalfindingswereobtainedforsolubleH-YantigenandMIS,i.e.,secretionbytesticularSertoliand,toalesserdegree,ovariancells,bindingtoagonad-specificreceptor,inductionofgonadalsexreversalinvitroand,incattle,invivo.H-Yantiseraalsodetectamoleculeormoleculesassociatedwiththeheterogameticsexinnon-mammalianvertebrates.Moleculardataonthisantigenorantigensarenotyetavailable.
