Fibroblastgrowthfactor16(FGF16)belongstothelargeFGFfamilywhichhasatleast23members.AllFGFfamilymembersareheparinbindinggrowthfactorswithacore120aminoacid(aa)FGFdomainthatallowsforacommontertiarystructure.FGFsareexpressedduringembryonicdevelopmentandinrestrictedadulttissues.Theyactoncellsofmesodermalandneuroectodermalorigintoregulatediversephysiologicfunctionsincludingangiogenesis,cellgrowth,patternformation,embryonicdevelopment,metabolicregulation,cellmigration,neurotrophiceffectsandtissuerepair.SignalingreceptorsforFGFsaretypeItransmembranereceptortyrosinekinasesbelongingtotheimmunoglobulin(Ig)superfamily.FourdistinctbutrelatedclassesofFGFreceptors,FGFR1,2,3,and4,exist.Throughalternativesplicing,multipleisoformsforFGFR1,2and3,withdistinctligandrecognitionprofiles,arealsogenerated.HumanFGF16cDNApredictsa207aaprecursorproteinwithoneN-linkedglycosylationsite.FGF16lacksatypicalsignalpeptidebutisefficientlygeneratedbymechanismsotherthantheclassicalproteinsecretionpathway.AmongFGFfamilymembers,FGF16ismostsimilartoFGF9,sharing73%aasequencehomology.HumanFGF16shares99%and98.6%aasequenceidentitywiththemouseandratFGF16,respectively.Inratembryos,FGF16messageisexpressedpredominantlyinbrownadipocytes.Inadultanimals,itislocalizedprimarilyinhearttissue.FGF16bindstoandactivatesFGFreceptor4.FGF16inducesproliferationofprimaryadipocytesandoligodendrocytesinvitroandstimulatesliverweightincreaseinvivo.TheexpressionpatternofFGF16anditseffectonadipocyteproliferationsuggestaroleforthisproteinontheproliferationofembryonicbrownadiposetissue.
