Podoplanin,alsoknownasglycoprotein36(gp36),PA2.26antigen,T1alpha(T1A),andaggrus,isa36kDatypeItransmembranesialoglycoproteinandmemberofthePodoplaninfamily.Podoplaninhasthreepotentialsplicevariants,thelongestofwhichisrepresentedbya238aminoacid(aa)precursor(NP_006465).Itcontainsanundefinedsignalsequence,a22aatransmembranesegment(aa207-228)andashortcytoplasmictail(aa229-238).TheECDcontainsabundantSer/ThrresiduesthatcouldserveaspotentialO-linkedglycosolationsites.ThecytoplasmictailcontainsputativesitesforproteinkinaseCphosphorylation.TherearetwopotentialalternatestartsitesatMet77(SwissProt#:Q86YL7)andMet119(EAW51692)thatgenerateshortforms.The162aashortformPodoplaninprecursorshares47%aaidentitywithmousePodoplanin.Podoplaninisexpressedonglomerularepithelialcells(podocytes),typeIlungalveolarcells,lymphaticendothelialcells,andnumeroustumors,includingcolorectaltumors,squamouscellcarcinomas,testicularseminoma,andbraintumors.OnestudyshowshighexpressionofPodoplaninmRNAinplacenta,lung,skeletalmuscle,andheart,andweakerlevelsinbrain,kidney,andliver.PodoplaninistheligandforC-typelectin-likereceptor2(CLEC2).TheirassociationisdependentonsialicacidonO-glycansofPodoplanin.ThroughitsassociationwithCLEC2,Podoplanin-inducesplateletaggregationandtumormetastasis.Podoplaninisalsonecessaryforlymphaticvesselformation,normallungcellproliferationandalveolusformationatbirth.
