Description:

EphA3,alsoknownasCek4,Mek4,Hek,Tyro4,andHek4,isamemberoftheEphreceptorfamilywhichbindsmembersoftheephrinligandfamily.Therearetwoclassesofreceptors,designatedAandB.BoththeAandBclassreceptorshaveanextracellularregionconsistingofaglobulardomain,acysteine-richdomain,andtwofibronectintypeIIIdomains,followedbythetransmembraneregionandcytoplasmicregion.Thecytoplasmicregioncontainsajuxtamembranemotifwithtwotyrosineresidues,whicharethemajorautophosphorylationsites,akinasedomain,andaconservedsterilealphamotif(SAM)inthecarboxytailwhichcontainsoneconservedtyrosineresidue.Activationofkinaseactivityoccursafterligandrecognitionandbinding.EphA3hasbeenshowntobindephrinA5,ephrinA2,ephrinA3,ephrinA1,ephrinA4,andephrinB1.TheextracellulardomainsofmouseandhumanEphA3sharegreaterthan96%aminoacididentity.OnlymembraneboundorFcclusteredligandsarecapableofactivatingthereceptorinvitro.Whilesolublemonomericligandsbindthereceptor,theydonotinducereceptorautophosphorylationandactivation.Invivo,theligandsandreceptorsdisplayreciprocalexpression.Ithasbeenfoundthatnearlyallreceptorsandligandsareexpressedindevelopingandadultneuraltissue.TheEph/ephrinfamiliesalsoappeartoplayaroleinangiogenesis.
