Description:

IL-13isa17kDaimmunoregulatorycytokinethatplaysakeyroleinthepathogenesisofallergicasthmaandatopy.ItissecretedbyTh1andTh2CD4+Tcells,NKcells,visceralsmoothmusclecells,eosinophils,mastcells,andbasophils.IL-13circulatesasamonomerwithtwointernaldisulfidebondsthatcontributetoabundledfouralpha-helixconfiguration.MaturehumanIL-13shares57%,59%,and94%aminoacidsequenceidentitywithmouse,rat,andrhesusIL-13,respectively.Despitethelowhomology,itexhibitscross-speciesactivitybetweenhuman,mouse,andrat.IL-13hasdiverseactivitiesonnumerouscelltypes.Onmacrophages,IL-13suppressestheproductionofproinflammatorycytokinesandothercytotoxicsubstances.OnBcells,IL-13inducesimmunoglobulinclassswitchingtoIgE,upregulatestheexpressionofMHCclassII,CD71,CD72,andCD23,andcostimulatesproliferation.IL-13upregulatesIL-6whiledownregulatingIL-1andTNF-alphaproductionbyfibroblastsandendothelialcells.IL-13bindswithlowaffinitytoIL-13Ralpha1,triggeringIL-13Ralpha1associationwithIL-4Ralpha.Thishighaffinityreceptorcomplexalsofunctionsasthetype2IL-4receptorcomplex.Additionally,IL-13bindswithhighaffinitytoIL-13Ralpha2whichisexpressedintracellularly,onthecellsurface,andasasolublemolecule.IL-13Ralpha2regulatesthebioavailabilityofbothIL-13andIL-4andisoverexpressedingliomaandseveralbronchialpathologies.ComparedtowildtypeIL-13,theatopy-associatedR110QvariantofIL-13elicitsincreasedresponsivenessfromeosinophilsthatexpresslowlevelsofIL-13Ralpha2.
