FormerlyknownasWnt-15orWnt-14b,Wnt-9bisasecretedglycoproteinbelongingtotheWntfamilyofsignalingproteinsthatarecriticallyinvolvedinmaintainingtheintegrityofbothembryonicandadulttissues.Wnt-9bisprimarilyexpressedinadultkidneysandduringlateembryogenesis,andshareswithotherWntfamilymembersthesamehighlyconservedlipid-modified,cysteine-richdomainessentialforcellsignaling.AsistrueformostWntfamilymembers,Wnt-9bfunctionsthroughthebiochemicalprocessknownasthecanonicalWntpathway;duringwhichWntproteinsbindtoandactivateseven-passtransmembranereceptorsoftheFrizzledfamily,andultimatelyresultinthedisruptionofbeta-catenindegradation.Intracellularaccumulationofbeta-cateninincreasestranslocationoftheproteinintothenucleus,whereitbindstoTCF/LEFtranscriptionfactorstopromotetheexpressionofnumerousgenes.Inthismanner,Wntsignalinginducesandmaintainstransformedphenotype,and,incertainembryoniccelllines,supportsself-renewalintheabsenceofsignificantdifferentiation.WhileincreasedWnt/beta-cateninsignalingisassociatedwithtumorigenesisinadiversesetofhumancancers,lackofWntsignalingdisruptstranscriptionalactivationoftumorsuppressorgenes,andhasbeenshowntoresultinneoplastictransformation,oncogenesis,andhumandegenerativediseases.AlteredWnt-9bexpressionhasbeenshowntoresultintheunderdevelopmentofthekidneys,andincompletelipandcleftfusioninmice.RecombinantHumanWnt-9bisamonomericglycoproteincontaining335aminoacidresiduesandhasacalculatedmolecularweightof36.9kDa.Duetoglycosylation,RecombinantHumanWnt-9bmigratesatanapparentmolecularweightofapproximately49-54kDabySDS-PAGEanalysisundernon-reducingconditions.
