Description:

Endothelialcellsexpressthreedifferentvascularendothelialgrowthfactor(VEGF)receptors,belongingtothefamilyofreceptortyrosinekinases(RTKs).TheyarenamedVEGFR-1(Flt-1),VEGFR-2(KDR/Flk-1),VEGFR-3(Flt-4).Theirexpressionisalmostexclusivelyrestrictedtoendothelialcells,butVEGFR-1canalsobefoundonmonocytes,dendriticcellsandontrophoblastcells.Theflt-1genewasfirstdescribedin1990.Thereceptorcontainssevenimmunoglobulin-likeextracellulardomains,asingletransmembraneregionandanintracellularsplitedtyrosinekinasedomain.ComparedtoVEGFR-2theFlt-1receptorhasahigheraffinityforVEGFbutaweakersignalingactivity.VEGFR-1thusleadsnottoproliferationofendothelialcells,butmediatessignalsfordifferentiation.InterestinglyanaturallyoccuringsolublevariantofVEGFR-1(sVEGFR-1)wasfoundinHUVEsupernatantsin1996,whichisgeneratedbyalternativesplicingoftheflt-1mRNA.
