Description:

CCL4,alsoknownasmacrophageinflammatoryprotein1beta(MIP1β),isa12kDaβchemokinethatissecretedatsitesofinflammationbyactivatedleukocytes,lymphocytes,vascularendothelialcells,andpulmonarysmoothmusclecells(1,2).CCL4attractsavarietyofimmunecellstositesofmicrobialinfectionaswellastootherpathologicinflammationsuchasallergicasthmaandischemicmyocardium.ACCL4deficiencyinmicepromotesthedevelopmentofautoantibodies,possiblyasaresultofcompromisedregulatoryTcellrecruitment.CCL4issecretedfromactivatedmonocytesasaheterodimerwithCCL3/MIP1α.ThefirsttwoN-terminalaminoacidscanbecleavedfromhumanCCL4byCD26/DPPIV.BoththefulllengthandtruncatedformsexertbiologicalactivitythroughCCR5,andthetruncatedformadditionallyinteractswithCCR1andCCR2.Inhumans,theabilityofCCL4tobindCCR5inhibitsthecellularentryofMtropicHIV1whichutilizesCCR5asacoreceptor.BothformsofCCL4blockHIV1infectionofTcellsbyinducingthedownregulationofCCR5.MaturemouseCCL4shares77%and86%aminoacidsequenceidentitywithhumanandratCCL4,respectively.
