Description:

Tumornecrosisfactoralpha(TNF-α),alsoknownascachectinandTNFSF1A,istheprototypicligandoftheTNFsuperfamily.Itisapleiotropicmoleculethatplaysacentralroleininflammation,apoptosis,andimmunesystemdevelopment.TNF-αisproducedbyawidevarietyofimmuneandepithelialcelltypes.HumanTNF-αconsistsofa35aminoacid(aa)cytoplasmicdomain,a21aatransmembranesegment,anda177aaextracellulardomain(ECD).WithintheECD,humanTNF-αshares97%aasequenceidentitywithrhesusand71%-92%withbovine,canine,cottonrat,equine,feline,mouse,porcine,andratTNF-α.The26kDatype2transmembraneproteinisassembledintracellularlytoformanoncovalentlylinkedhomotrimer.Ligationofthiscomplexinducesreversesignalingthatpromoteslymphocytecostimulationbutdiminishesmonocyteresponsiveness.
