Description:

Thrombospondins(TSP)aresecretedmultidomainglycoproteinswithmanyputativefunctionsincludingmodulatingcelladhesion,proliferation,migration,andangiogenesis.AtleastfiveTSPsexist:TSP-1,-2,-3,-4,andcartilageoligomericglycoprotein(COMP)/TSP-5.Thefamilycanbedividedintotwosubgroups.Thefirst,TSP-1and-2,arehomotrimericmolecules,eachmonomercontaininganN-terminal/heparin-bindingdomain,acoiled-coiloligomerizationdomain,avonWillebrandfactor-type/procollagenhomologydomain,threetype1thrombospondinrepeats(TSR),threeEGF-likerepeats,sevenTSPtype3repeats,andaC-terminalglobulardomain.TSP-1and-2arethoughttomediateinteractionsbetweencellsandtheextracellularmatrix,andassucharematricellularproteins.Incontrast,TSP-3,-4,andCOMP/TSP-5arehomopentamersthatlacktheN-terminalstructureofTSP-1and-2,buthaveasimilararrangementofEGFandTSPtype3repeats,andtheC-terminaldomaincharacteristicoftheTSPfamily.
