Description:

RecombinanthumansolubleVascularEndothelialGrowthFactorReceptor-1(sVEGFR-1)isthenaturallyoccurringformandwasclonedfromtotalRNAofhumanumbilicalveinendothelialcells.TherecombinantmaturesVEGFR-1isaglycosylatedmonomericproteinwithamassofapproximately96kDa.Thesolublereceptorproteinconsistsofthefirst6extracellulardomains(Met1-His688)containingtheunique31aminoacidsresiduesattheC-terminus.Endothelialcellsexpressthreedifferentvascularendothelialgrowthfactor(VEGF)receptors,belongingtothefamilyofreceptortyrosinekinases(RTKs).TheyarenamedVEGFR-1(Flt-1),VEGFR-2(KDR/Flk-1),andVEGFR-3(Flt-4).Theirexpressionisalmostexclusivelyrestrictedtoendothelialcells,butVEGFR-1canalsobefoundonmonocytes,dendriticcellsandontrophoblastcells.Theflt-1genewasfirstdescribedin1990.Thereceptorcontainssevenimmunoglobulin-likeextracellulardomains,asingletransmembraneregionandanintracellularsplittyrosinekinasedomain.ComparedtoVEGFR-2theFlt-1receptorhasahigheraffinityforVEGFbutaweakersignalingactivity.VEGFR-1thusleadsnottoproliferationofendothelialcells,butmediatessignalsfordifferentiation.Interestingly,anaturallyoccurringsolublevariantofVEGFR-1(sVEGFR-1)wasfoundinHUVECsupernatantsin1996,whichisgeneratedbyalternativesplicingoftheflt-1mRNA.ThebiologicalfunctionsofsVEGFR-1stillarenotclear,butitseemstobeanendogenousregulatorofangiogenesisbindingVEGFwiththesameaffinityasthefull-lengthreceptor.
