Description:

RecombinanthumansolubleVascularEndothelialGrowthFactorReceptor-1domainD1-5(sVEGFR-1(D5))isa70kDaprotein.Thebaculovirusgenerated,recombinanthumansVEGFR-1isproducedasanon-chimericproteininamonomericform.Thesolublereceptorproteincontainsonlythefirst5extracellulardomains,whichcontainalltheinformationnecessaryforhighaffinityligandbinding.Thereceptormonomershaveamassofapproximately70kDa.Endothelialcellsexpressthreedifferentvascularendothelialgrowthfactor(VEGF)receptors,belongingtothefamilyofreceptortyrosinekinases(RTKs).TheyarenamedVEGFR-1(Flt-1),VEGFR-2(KDR/Flk-1),VEGFR-3(Flt-4).Theirexpressionisalmostexclusivelyrestrictedtoendothelialcells,butVEGFR-1canalsobefoundonmonocytes,dendriticcellsandontrophoblastcells.Theflt-1genewasfirstdescribedin1990.Thereceptorcontainssevenimmunoglobulin-likeextracellulardomains,asingletransmembraneregionandanintracellularsplitedtyrosinekinasedomain.ComparedtoVEGFR-2theFlt-1receptorhasahigheraffinityforVEGFbutaweakersignalingactivity.VEGFR-1thusleadsnottoproliferationofendothelialcells,butmediatessignalsfordifferentiation.InterestinglyanaturallyoccuringsolublevariantofVEGFR-1(sVEGFR-1)wasfoundinHUVECsupernatantsin1996,whichisgeneratedbyalternativesplicingoftheflt-1mRNA.ThebiologicalfunctionsofsVEGFR-1stillarenotclear,butitseemstobeanendogenousregulatorofangiogenesis,bindingVEGFwiththesameaffinityasthefull-lengthreceptor.
